SCREENING OF BASIC IMMUNOLOGICAL MARKERS

The starting point for precise leukocyte phenotyping is identifying the frequency of basic immune cell subpopulations. The prepared panel is a classical yet slightly extended TBNK test, allowing simultaneous analysis of major T cell groups (CD3+, CD4+, CD8+), B cells (CD19+), and NK cells (CD16+/CD56+). The addition of markers such as CD14 enables the evaluation of monocyte presence and function, while CD45 allows for an accurate determination of the total leukocyte count. The extended panel provides a broad range of information on the composition and proportions of various immune cell subpopulations.
This analysis allows for the rapid and precise identification of immune system dysfunctions, which is particularly useful in diagnosing autoimmune diseases, hematological malignancies (e.g., leukemias, lymphomas), and monitoring blood changes in patients undergoing immunosuppressive or immunostimulatory therapies. It is also a key tool in monitoring the course of viral infections, such as HIV, where changes in CD4+ and CD8+ lymphocyte subpopulations can serve as indicators of disease progression.

THE STORY OF „T”

The panel is perfectly suited for screening the activation status and maturity of T cells.

  1. Naive T cells (CD3+ CD4+ or CD8+, CD45RA+, CCR7+) – Cells that have not yet encountered an antigen.

  2. Central memory T cells (TCM, CD3+ CD4+ or CD8+, CD45RO+, CCR7+) – Memory cells that migrate to lymph nodes and are capable of providing a long-term response to antigens.

  3. Effector memory T cells (TEM, CD3+ CD4+ or CD8+, CD45RO+, CCR7-) – Effector memory cells found in tissues that can immediately respond to the reappearance of an antigen.

  4. Terminally differentiated effector T cells (TEMRA, CD3+ CD8+, CD45RA+, CCR7-) – Highly cytotoxic CD8+ T cells that have completed differentiation.

  5. Senescent T cells (CD57+, CD28-) – T cells that have lost their ability to proliferate and often show characteristics of aging.

  6. Exhausted T cells (PD-1+) – T cells that have been chronically activated and may be functionally impaired, often seen in chronic infections or cancers.

Treg Research Panel

    This panel can be used to monitor changes in Treg populations and their functions in diseases like rheumatoid arthritis, multiple sclerosis, type 1 diabetes, or systemic lupus erythematosus.

  1. Cancer Immunotherapy:

    Analyzing Treg populations is crucial in the context of cancers, where excessive Treg activity can suppress the anti-tumor response. Monitoring these cells during immunotherapies can help assess treatment effectiveness.
