REAGENTS FOR THE MAINTENANCE OF CYTOMETERS AND CELL SORTERS
Proper maintenance of flow cytometers and cell sorters primarily requires regular rinsing and/or cleaning of the instrument’s fluidics and pressure systems. The most critical aspect of these maintenance activities is preventing the formation of microbial biofilms in the capillaries and tubing, as well as the precipitation of salts from the sheath fluid.
Equally important is the implementation of quality control (QC) practices, which are part of good cytometry practices. QC in flow cytometry is essential to ensure accurate, reliable, and reproducible data from experiments. It encompasses a series of practices, protocols, and controls designed to verify that the flow cytometer operates optimally and that experimental results are consistent over time and across different devices. QC includes:
Calibration and standardization of instruments,
Monitoring instrument functions (sensitivity, stability, and compensation settings over time),
Fluorescence compensation,
Quality control of antibodies and reagents,
Standardization of data acquisition protocols,
Data analysis monitoring (result reproducibility and the creation of standard operating procedures (SOP) for each application),
Monitoring correct instrument usage and troubleshooting.
